I will briefly describe the fundamentals of both methods\, their technol ogical implementation\, hybrid probes and the developments that led from in-vivo studies on experimental rodent models ("bench-top") to pilot cl inical studies ("bed-side"). I will discuss potential applications and l imitations of this technology. Photon scattering offers information about the structural aspects of a tissue. Architectural landmarks are imaged by r eflectance from refractive index mismatches at inter-layer interfaces\, eg.\, OCT imaging of multi-layered tissues. Ultrastructural characteriz ation of the tissue between such interfaces can be documented by (1) the depth dependence of attenuation of backscattered reflectance signal (R) \, (2) the wavelength dependence of R\, and (3) the angular dependence o f R. If thin samples are used\, characterization can utilize transmissi on measurements (T). The interpretation of such R and T measurements re quires an accurate model of the size\, shape and number density distribu tions of scattering particles in a tissue\, or the equivalent continuum description as the spatial frequency of refractive index fluctuations. Usually\, such a model is lacking. Nevertheless\, a fingerprint of chara cterization can be derived using simple Mie theory to describe an equiva lent solution of microspheres that mimics the observed scattering behavi or of a tissue. This webinar discusses the issues involved in characteri zing a tissue by scattering. The goal is a label-free contrast mechanism for imaging cancer margins to guide surgery\, and as a method of follow ing changes in tissues induced by pathology or drug treatments.\n\nSpeak er Bio:\nSteven L. Jacques\, Ph.D.\, received a B.S. degree in Biology a t M.I.T.\, and an M.S. degree in Electrical Engineering and Computer Sci ence and a Ph.D. degree in Biophysics and Medical Physics from the Unive rsity of California-Berkeley (1984)\, where he used dielectric microwave measurements to explore the in vivo distribution of water in the stratu m corneum of human skin. His postdoctoral work was at the Wellman Cente r for Photomedicine at Massachusetts General Hospital\, rising to the po sition of Lecturer in Dermatology/Bioengineering\, Harvard Medical Schoo l. His team developed the use of Monte Carlo computer simulations to stu dy optical transport in biological tissues\, which is now widely used in the field of biophotonics. \nIn 1988\, he joined the University of Tex as M. D. Anderson Cancer as an Assistant Professor of Urology/Biophysics and established a laboratory developing novel laser and optical methods for medicine\, later achieving a tenured position as Associate Professo r. He developed a hand-held spectrometer and the analysis software to no ninvasively measure hyperbilirubinemia in newborns. This device was pate nted\, licensed\, and FDA approved to replace painful heel stick tests\, and is now in practice in neonatal care. As of 2009\, over 20 million n ewborns have been tested with the device. In 1996\, he joined the Oregon Health & Science University in Portland\, Oregon\, where he now serves as Professor of Dermatology and Biomedical Engineering. His work contin ues on developing novel uses of optical technologies for both therapy an d diagnosis. Currently\, he has developed a hand-held polarized light ca mera to visualize skin cancer margins and guide surgical excision\, now in clinical trials. He has developed in vivo sub-nm measurements of vibr ation of the cochlear membrane of the inner ear in animal models. He is developing novel microscopes that are sensitive to the ultrastructure of cells and tissues. To understand the neuronal function\, it is essential observ e the firing patterns of neuronal population with high spatiotemporal re solution Based on the dependency relationship between neuronal spiking a nd the corresponding calcium signal\, the firing pattern have been monit ored with sub-cell spatial resolution using two-photon imaging. However\ , it is still a big challenge to acquire the fast activation pattern of the neuronal populations\, and assure their dynamic functional connectio ns. Here we show 1) the development of a two-photon imaging system with an inertia-free scanning technique to acquire the fast activity of neuro nal populations\, and 2) a method to reconstruct the activation pattern with single spike temporal resolution\, and thus are able to monitor the dynamic functional connections. With the high spatio-temporal resolutio n\, this method is expected to be a starting point to spy the function o f the microcircuits\, such as information encoding and signal processing .\n\nBIO\n\nDr. Shaoqun Zeng is a HUST Distinguished Professor\, Deputy Director of the Britton Chance Center for Biomedical Photonics\, Directo r of the Division of Biomedical Photonics\, at Wuhan National Laboratory for Optoelectronics\, and Vice-Dean for Research at the Huazhong Univer sity of Science and Technology (HUST). \nHis research interests include femtosecond laser biomedical imaging\, new methods in high resolution op tical functional imaging of proteins and biomedical modelling. In the early days visualizing the structure of living cells lead to enormous insights. More recently the emphasis of optical imaging has shifted from structural to functiona l imaging\, where very specific biological questions can be addressed at microscopic and\, in some cases\, nanoscopic scales. The importance of functional imaging has\, of course\, been recently recognized by the awa rd of the 2008 Nobel Prize for Chemistry for the developments in Green F luorescent Protein as a unique means to spatially locate processes in ce lls.\n\nFunctional imaging is a highly interdisciplinary area bringing t ogether biologists\, chemists\, physicists and engineers. Our institute of Biophysics\, Imaging and Optical Science has recently been establishe d to bring together scientists in these disciplines and the present meet ing is the inaugural conference representing this field.\n\nWe are fortu nate to have attracted an extremely eminent range of speakers across man y disciplines who will provide exciting visions of their own research wh ich we expect to stimulate lively discussions leading to new research di rections. \n\nPlaces are limited.
